SYP Antibody (C-12) is a mouse monoclonal IgG1 antibody that detects synaptophysin (SYP) in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). SYP is an integral membrane protein predominantly found in neurons and endocrine cells, playing a crucial role in the formation and recycling of synaptic vesicles. SYP is characterized by N-glycosylation and the ability to form hexameric complexes, which are essential for function as a large conductance channel. SYP contains four transmembrane domains, which are vital for structural integrity and functionality, allowing facilitation of neurotransmitter transport. SYP interacts with membrane cholesterol, which is critical for vesicle formation, and forms complexes with synaptobrevin (VAMP) that are upregulated during neuronal development. While the synaptophysin-synaptobrevin complex is not essential for the exocytosis process, this complex serves as a reservoir of synaptobrevin, ensuring sufficient levels are available for neurotransmitter release. Additionally, the tail domain of brain Myosin V forms a stable complex with synaptobrevin II and SYP, which disassembles upon calcium influx during neuronal depolarization, highlighting the dynamic nature of these interactions in synaptic function. SYP Antibody (C-12) serves as an invaluable tool for researchers studying synaptic mechanisms and synaptophysin′s role in neurobiology.
