SYP Antibody (A-9) is a mouse monoclonal IgG2b antibody that detects synaptophysin (SYP) in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). SYP is an integral membrane protein predominantly found in neurons and endocrine cells, playing a crucial role in the regulation of synaptic vesicle dynamics. SYP forms hexameric complexes that contribute to the formation of large conductance channels, which are essential for neurotransmitter release and synaptic transmission. SYP contains four transmembrane domains and is N-glycosylated, which is important for stability and function. SYP interacts with membrane cholesterol, facilitating vesicle formation and recycling, a process vital for maintaining synaptic function. SYP forms complexes with synaptobrevin (VAMP), which are upregulated during neuronal development, although these complexes are not essential for the exocytosis process itself. These complexes serve as a reservoir of synaptobrevin, ensuring a readily available supply for neurotransmitter release. Dynamic interactions of SYP with other proteins, such as brain Myosin V, further underscore SYP′s importance in synaptic physiology, particularly during calcium-mediated signaling events that trigger neurotransmitter release.
