
Syk Antibody (G-2) is a mouse monoclonal IgG2a antibody that detects Syk in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Syk, or spleen tyrosine kinase, is a 635 amino acid protein characterized by one protein kinase domain and two SH2 domains, playing a crucial role as a positive effector in B cell antigen receptor (CD79)-stimulated responses. Syk is essential for coupling CD79 with calcium ion movement through two phospho-regulated pathways, which are vital for various cellular processes. Specifically, when unphosphorylated, Syk directs calcium ions through a phosphoinositide 3-kinase-dependent pathway, while phosphorylation at Tyr 348 and Tyr 352 shifts the pathway to a phospholipase C γ-dependent route. Syk regulation of calcium movement influences critical cellular responses such as differentiation, phagocytosis, proliferation, and B cell development. Additionally, Syk expression shows notable upregulation in T cell lymphoma, indicating potential involvement in tumorigenesis.
