Syk Antibody (D-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Syk in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Syk (D-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Syk, or spleen tyrosine kinase, is a 635 amino acid protein characterized by one protein kinase domain and two SH2 domains, playing a crucial role in B cell signaling. Specifically, Syk acts as a positive effector in B cell antigen receptor (CD79)-stimulated responses, facilitating calcium ion movement through two distinct phospho-regulated pathways. When Syk is unphosphorylated, calcium ions travel via a phosphoinositide 3-kinase-dependent pathway, while phosphorylation at tyrosine residues 348 and 352 shifts calcium flow to a phospholipase C γ-dependent pathway. This regulation of calcium movement is vital for various cellular processes, including differentiation, phagocytosis, proliferation, and B cell development. Notably, Syk expression is upregulated in T cell lymphoma, indicating potential involvement in tumorigenesis. Additionally, Syk exists in two isoforms, short and long, resulting from alternative splicing events, which may contribute to functional diversity in cellular signaling pathways.
