SV2A Antibody (E-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects SV2A protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-SV2A antibody (E-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SV2A protein, also known as Synaptic Vesicle 2A, plays a crucial role in synaptic transmission by regulating cytoplasmic calcium levels in presynaptic nerve terminals during repetitive stimulation, which is essential for maintaining normal brain function. SV2A protein is predominantly expressed in the subcortex and is found on all synaptic vesicles, regardless of the type of neurotransmitter, highlighting its importance in the overall process of neurotransmission. Additionally, SV2A is functionally redundant with SV2B, another isoform, ensuring that synaptic transmission remains efficient even if one isoform is compromised. The differential expression of SV2A and SV2B in the brain underscores their specific roles in various synaptic functions, making SV2A (E-8) monoclonal antibody a valuable tool for researchers studying synaptic mechanisms and neurobiology.

Thank you for your question. SV2A Antibody (E-8): sc-376234 is supplied as 200 µg in 1 ml PBS with 0.1 % gelatin and <0.1 % sodium azide. It is is recommended for detection of SV2A of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. It is also reactive with additional species, including and equine, canine, bovine and porcine samples.

With antigen retrieval buffer was used to test this antibody on human tissue?

Product codes sc-376234 FITC, sc-376234 PE, sc-376234 AF488, sc-376234 AF546, sc-376234 AF594, sc-376234 AF647, sc-376234 AF680, and sc-376234 AF790 are supplied as fluorescent direct conjugates. The corresponding fluorochromes are bound to the antibody, which means that no secondary antibody is required for detection.

Following incubation with beads coated in anti-SV2A, IP pellet, supernatant, and final wash samples were fractionated by SDS-PAGE and Western blotted onto a nitrocellulose membrane and detected using an anti-SV2A rabbit polyclonal Ab from a different supplier. No SV2A was detected in either supernatant or final wash samples, demonstrating that this Ab has a high affinity for SV2A, and is ideal for IP.

Rated 5 out of 5 by from Excellent marker for SV2AWe obtained a sample of SV2A monoclonal from SCB after difficulties with the same antibody from different providers. We used this antibody in IHC-P for calculating synaptic density in human brain tissue. The staining was incredibly consistent between samples, very easily quantifiable and led to a publication. We will continue using this marker from SCB. Very satisfied.
