SREBP-1 Antibody (E-4) is a mouse monoclonal IgG2b kappa light chain antibody that detects SREBP-1 protein of mouse, rat, and human origin through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry (IHC), and enzyme-linked immunosorbent assay (ELISA). SREBP-1 (E-4) antibody is offered in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. SREBP-1 protein plays a crucial role in lipid metabolism by regulating the expression of genes involved in cholesterol and fatty acid synthesis. SREBP-1 protein is synthesized as a precursor that resides in the nuclear envelope and endoplasmic reticulum, and upon sterol depletion, undergoes proteolytic cleavage to release a soluble fragment that translocates to the nucleus, where SREBP-1 activates transcription of target genes, including the low-density lipoprotein receptor. This receptor is essential for the endocytic uptake of cholesterol-carrying lipoproteins, thereby maintaining cellular and plasma cholesterol levels. SREBP-1 regulation is vital for cellular homeostasis, as excessive cholesterol can lead to cellular dysfunction and contribute to diseases such as atherosclerosis.

Does this antibody identifies both mature and immature forms of SREBP1?

Thank you for your question. This mouse monoclonal antibody was raised against amino acids 41-200 of SREBP-1 of human origin, but the antibody has not been epitope mapped so further information regarding the epitope location is not available at this time. In Western blot experiments the precursor form is most commonly detected with this antibody.

When should the TransCruz reagent sc-17755 X be used instead of the standard form sc-17755?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

Supposed size should be 125 (135) KD, but H1299 is about 150KD, and A549 have light band about 200KD? Not consider A549 can visualize bands. H1299 have a light band at around 80KD. Not sure what are they
