SPT6 Antibody (E-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects SPT6 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SPT6 Antibody (E-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SPT6 protein, also known as suppressor of Ty6 homolog, SUPT6H, SPT6H, or Tat-cotransactivator 2 protein, is a highly conserved 1,726 amino acid protein that plays a crucial role in regulating transcriptional elongation by RNA polymerase II (Pol II). Located predominantly in the nucleus, SPT6 contains one SH2 domain and one S1 domain, which are essential for interaction with other nuclear proteins. Notably, SPT6 interacts with SPT4 and SPT5, forming the DRB-sensitivity-inducing factor (DSIF) complex that binds to RNA polymerase II and directly influences transcriptional elongation. Additionally, SPT6′s C-terminus allows interaction with Histone H3, further emphasizing SPT6′s role in chromatin dynamics and gene expression regulation. Three isoforms of SPT6, resulting from alternative splicing, highlight the complexity of function and regulation in various cellular contexts.
