SPT5 Antibody (D-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects SPT5 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). SPT5 (D-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SPT5, also known as DSIF p160, plays a crucial role in regulating transcriptional elongation, particularly in the context of HIV-1 protein Tat and DRB-mediated transcriptional inhibition. SPT5 forms a complex with SPT4, known as DRB-sensitivity-inducing factor (DSIF), which interacts with RNA polymerase II to modulate transcription elongation. SPT5 undergoes post-translational modifications, particularly phosphorylation, which significantly affects function. In mitotic HeLa cells, SPT5 exhibits slower migration on SDS-PAGE compared to interphase counterpart due to enhanced phosphorylation, particularly at C-terminal CTR1 domain, which is a substrate for P-TEFb phosphorylation. This modification is essential for SPT5′s role as a transcriptional elongation regulator, highlighting the importance of post-translational modifications in functional dynamics.
