SPT4 Antibody (A-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects SPT4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SPT4 (A-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SPT4, also known as suppressor of Ty4 and p14, plays a crucial role in transcriptional elongation regulation, particularly in HIV-1 protein Tat and DRB (5,6-dichloro-1-beta-D-ribofuranosylbenzimidazole)-mediated transcriptional inhibition. SPT4 forms a complex with SPT5, known as DRB-sensitivity-inducing factor (DSIF), which interacts with RNA polymerase II to modulate transcription elongation. Notably, SPT5 phosphorylation, particularly in mitotic HeLa cells, alters migration on SDS-PAGE, indicating a dynamic regulatory mechanism essential for function. P-TEFb complex specifically phosphorylates SPT5′s C-terminal CTR1 domain, which is vital for transcriptional elongation regulation. Additionally, murine homologs Supt4h1 and Supt4h2 contribute to mRNA processing and transcription elongation, highlighting SPT4′s importance in gene expression regulation across species.
