Sp3 Antibody (F-11) is a mouse monoclonal IgG2a light chain antibody that detects Sp3 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Sp3 is a member of the Sp transcription factor family, which also includes Sp1, Sp2, and Sp4, and is characterized by a highly conserved DNA-binding domain featuring three zinc fingers at its C-terminus. This structural feature allows Sp3 to bind to specific DNA sequences known as GC boxes, which are essential for the regulation of gene expression. Sp3 plays a unique role in transcriptional regulation of various genes by inhibiting the activity of other Sp family members, such as Sp1 and Sp4, thereby fine-tuning the transcriptional response in different cellular contexts. Sp3′s interaction with regulatory elements, such as the E2 element for the β4 subunit of neuronal nicotinic acetylcholine receptors, highlights Sp3′s importance in neuronal signaling and development. Multiple isoforms of Sp3, generated through alternative splicing, add another layer of complexity to Sp3′s functional repertoire, making Sp3 a critical player in cellular processes and a valuable target for research in gene regulation and neurobiology.
