Sos 2 (B-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects Sos 2 protein in mouse, rat and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF) and enzyme-linked immunosorbent assay (ELISA). Anti-Sos 2 antibody (B-6) is available unconjugated and in multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC) and various Alexa Fluor® conjugates. The superfamily of GTP-binding proteins, including Ras proteins as prototypes, plays roles in diverse biological activities. Research has identified guanine nucleotide-releasing factors (GRFs) that activate Ras in mammalian cells and an "adapter" protein (Sem 5/GRB2) that mediates GRF interactions with activated receptor molecules. Ras-GRF p140 promotes nucleotide exchange specifically on Ras p21s but not other Ras gene superfamily members. Three mammalian homologs of Drosophila Ras-GRF, son of sevenless (Sos), have been identified - mSos 1 and mSos 2 from mouse, and hSos from human. Vav p95 functions as a GRF in Ras activation by T cell receptor and contains a domain similar to Dbl p115, a GRF specific for Cdc42Hs. Following activation, Ras interacts with Raf to activate the MAP kinase phosphorylation pathway.

Rated 5 out of 5 by from clone B6 worked in a Western using HeLa and LN229I've been struggling to see a signal for SOS2 in HeLa lysates. A coworker tested several antibodies from other vendors and chose one that just wasn't very potent. I tested 3 SCBT antibodies (clones A8, B6, and G7) in a single experiment: HeLa cells vs LN229 cells, treated with an inhibitor or DMSO control and then stimulated with EGF for 5'; 40µg whole cell lysate (using NP40) per lane on a 3%-8% tris-acetate gel run at 85V for 1.2hrs, then turned up to 135V till the dye ran off the bottom. Protein transfer was to a single PVDF membrane in an iBlot2 with a custom program (created to transfer large proteins). After transfer, the blot was cut into 3 blocks. All blocks were blocked with 5% BSA in TBS for 1.5hrs, then Tween-20 was added to 0.1%, primary antibody was added at a 1:400 dilution, and the blots were incubated overnight in a cold room on a rocker. The following day, the blots were washed 4 times (5' each) with TBST, then incubated with Licor IR800 secondary antibody. Incubation was for 1.5hrs after which they were washed 4 times (5' each) and imaged using a Licor Odyssey. Only the B6 antibody showed banding. I've reviewed the other 2 antibodies separately.
