SEE ALSO...

SNAP 25 Antibody (SP12) is a mouse monoclonal IgG1 kappa light chain antibody that detects SNAP 25 protein of broad species origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-SNAP 25 antibody (SP12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SNAP 25 plays a crucial role in synaptic transmission as SNAP 25 is involved in the docking and fusion of synaptic vesicles with the presynaptic membrane, a process essential for neurotransmitter release. This function is vital for proper neuronal communication and overall brain function, as disruptions in SNAP 25 activity can lead to neurological disorders and impairments in synaptic plasticity. SNAP 25 interacts with several key proteins in the exocytosis pathway, including syntaxins and vesicle-associated membrane proteins (VAMPs), which together form a complex that facilitates the fusion of vesicles with the plasma membrane. Understanding SNAP 25 interactions and functions is important for elucidating the mechanisms underlying synaptic transmission and potential therapeutic targets for treating synaptic dysfunction.

Hi, Has Santacruz conducted any internal test/experiment for use of this antibody in ELISA? or any reference/citation for the same?

Asked by: ATGC
Thank you for your question. SNAP 25 Antibody (SP12): sc-20038 is recommended for detection of SNAP 25 of broad species origin by WB, IP, IF, IHC(P) and solid-state ELISA. It has been tested and established for these applications in our labs using our standard protocols. You can find our protocols by following this link: https://www.scbt.com/scbt/resources/protocols

Hello, has anyone tested this antibody on mouse frozen brain sections? Thanks in advance for your feedback.

Asked by: MarieV
Thank you for your question. This antibody has been published in mouse brain samples, and we do list mouse brain as a positive control for this antibody, so yes, I would say that this antibody is perfectly fine for IF use on mouse brain samples. If you have any further questions, please contact our European Technical Service team. You can reach them by phone at Toll Free: +49 6221 4503 0, by email at: europe@scbt.com or by live chat directly on our website, www.scbt.com

Is this SNAP 25 Antibody (SP12) suitable for detecting the cleaved product of SNAP25 by BoNT/A? Which region of SNAP25 is critical for this antibody to recognize SNAP25? Thanks a lot,

Asked by: John123
Thank you for your question. The exact epitope recognized by this mouse monoclonal antibody, SNAP 25 (SP12): sc-20038, has not been determined. And Santa Cruz Biotechnology has not conducted internal testing to evaluate detection of the cleaved SNAP 25 protein by BoNT/A. However, this antibody has been successfully used before for this purpose. Please refer to the following journal article on PubMed which cites the SP12 clone provided by Santa Cruz Biotechnology. Rated 3 out of 5 by from Works OK for western blots of SNAP25 in human cellThe SNAP 25 antibody showed the expected 25kDa band on western blots with the lysates of hTERT-RPE1 (human retinal epithelial) cells. The intensity of the 25 kDa band corresponded to mRNA levels measured by RNAseq. However, several apparently non-specific higher molecular weight bands showed up on the blots as well
