

Skeletrophin Antibody (D-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects Skeletrophin protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Skeletrophin antibody (D-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Skeletrophin, also known as MIB2 (Mindbomb homolog 2), SKD, ZZANK1, or ZZZ5, functions as an E3 ubiquitin-protein ligase that plays a crucial role in regulating Notch signaling through targeted degradation of specific proteins. This regulation is vital for maintaining proper cellular communication and differentiation, particularly in developmental processes and tissue homeostasis. Skeletrophin is predominantly expressed in heart, brain, kidney, and skeletal muscle, where Skeletrophin positively influences Notch signaling by ubiquitinating the intracellular domain of Delta receptors, which serve as ligands for Notch proteins. This ubiquitination process promotes transendocytosis of the Notch extracellular domain, facilitating transport of the Notch intracellular domain to the nucleus, where Skeletrophin activates a range of genes essential for various biological functions. Notably, Skeletrophin localizes to the endosome and exhibits down-regulation in primary skin melanomas, with studies indicating that treatment with 5′-aza-2-deoxycytidine, a demethylating agent, can enhance Skeletrophin expression, suggesting that down-regulation may be linked to methylation of the Skeletrophin gene. Additionally, six isoforms of Skeletrophin exist due to alternative splicing events, further underscoring the complexity and significance of this protein in cellular signaling pathways.
