    SEE ALSO...

Siglec-1 Antibody (B-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects Siglec-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Siglec-1 antibody (B-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Siglec-1, also known as CD169, plays a crucial role in the immune system as a sialic acid-binding receptor expressed on hemopoietic cells, particularly macrophages. Siglec-1 is involved in mediating cell-cell interactions within lymphoid tissues, which is essential for proper functioning of immune responses. Notably, Siglec-1 has a higher affinity for N-acetylneuraminic acid (Neu5Ac) compared to N-glycolylneuraminic acid (Neu5Gc), highlighting specificity in recognizing sialylated ligands. The human Siglec-1 gene is located on chromosome 20p13 and encodes a 1,709 amino acid protein, with alternative splicing resulting in a soluble isoform that is not membrane-bound. This unique structural characteristic allows Siglec-1 to participate in various immunological processes, including regulation of immune cell interactions and modulation of inflammatory responses, making Siglec-1 a significant target for research in immunology and related fields.

What blocking solution should I use with Siglec-1 (B-1): sc-398109 monoclonal antibody for Western blot?

    Asked by: Randy McDonald
Thank you for your question. We recommend blocking with Blotto (either TBS Blotto A: sc-2333 or TBS Blotto B: sc-2335). Our Western blot protocol can be found here: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting

Rated 5 out of 5 by from Western blotWe tested the expression of Siglec-1 protein in different groups of cells by Siglec-1 antibody,we optimized the conditions about the antibody dilutions and blocking conditions and transfer conditions. We use the beta actin as the internal control. The primary antibody 1:500 maybe a little better, recommend use BSA 5% for blocking buffer .We obtained the specific results and signal is in according with the sample group controls.
