SIGIRR Antibody (G-8) is a mouse monoclonal IgG1 antibody that detects SIGIRR in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SIGIRR, also known as single immunoglobulin domain-containing IL-1 receptor-related protein or Toll/interleukin-1 receptor 8 (TIR8), plays a crucial role as a negative regulator of the interleukin-1 receptor and Toll-like receptor signaling pathways, which are vital for the immune response. By modulating these pathways, SIGIRR helps to prevent excessive inflammation and tissue damage, thereby conferring resistance to infections, such as those caused by Pseudomonas aeruginosa in corneal tissues. SIGIRR is primarily localized in the kidney, lung, and gut, where SIGIRR expression is essential for maintaining immune homeostasis and protecting against pathogenic challenges. Additionally, SIGIRR can form complexes with key signaling molecules, including IL-1 receptor 1, MYD-88, IRAK-1, and TRAF-6, upon stimulation, highlighting SIGIRR′s integral role in the immune signaling cascade. With SIGIRR′s ability to interact with these proteins, SIGIRR serves as a critical checkpoint in the immune response, making SIGIRR a valuable target for research in inflammation and immune regulation.
