SARA Antibody (B-9) is a mouse monoclonal IgG2b kappa light chain antibody that detects SARA protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SARA (B-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SARA protein, also known as NSP, MADHIP, SMADIP, or ZFYVE9, features a double zinc finger FYVE domain that plays a crucial role in recruiting Smad proteins to the TGF-beta receptor, ensuring proper subcellular localization of the activated receptor-bound complex. The FYVE domain directs SARA to early endosomal compartments, where SARA interacts with TGF-beta receptors and Smads, facilitating effective signaling pathways. SARA physically interacts with the promyelocytic leukemia (PML) tumor suppressor, which enhances the association and accumulation of SARA and the TGF-beta receptor within early endosomes. SARA can modulate the recruitment of protein phosphatase 1 catalytic subunit (PP1c) to the Smad7-GADD34 complex, influencing the dephosphorylation of the TGF-beta receptor by Smad7, a critical mechanism for regulating negative feedback in TGF-beta signaling. This intricate interplay underscores SARA′s importance in maintaining cellular homeostasis and potential implications in various pathological conditions.
