
SAP 61 Antibody (G-4) is a mouse monoclonal IgG1 antibody that detects SAP 61 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). SAP 61, also known as SF3A3, PRP9, PRPF9, or SF3a60, is a 501 amino acid protein characterized by the presence of one matrin-type zinc finger and is a crucial component of the SF3A splicing factor complex. This complex, which is essential for pre-mRNA splicing, is localized in the nucleus and consists of three subunits that work together to facilitate the binding of U2 small nuclear ribonucleoprotein (snRNP) to the branchpoint sequence in pre-mRNA. SAP 61 functions as the third subunit of the SF3A complex, interacting with subunit 1 (SAP 114) through its N-terminus while simultaneously binding to the 15S U2 snRNP via its zinc finger domain. This interaction enables the conversion of the 15S U2 snRNP into the active 17S form, which is directly involved in pre-mRNA splicing events. SAP 61 plays an indispensable role in prespliceosome assembly and overall cell viability, highlighting its critical function in gene expression regulation. A pseudogene for SAP 61 has been identified on chromosome 20, underscoring the evolutionary significance of this protein.
