Sab Antibody (PL-A23) is a mouse monoclonal IgG1 antibody that detects Sab in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Sab is a Src homology 3 domain (SH3) binding protein that plays a critical role in regulating Bruton′s tyrosine kinase (Btk), a key player in B cell development. Anti-Sab antibody (PL-A23) recognizes the interaction between Sab and Btk, which serves as a negative regulatory mechanism; Sab binding to Btk inhibits kinase activity, thereby reducing phosphorylation of Btk substrates and preventing Btk-induced auto-phosphorylation in B cells. This regulatory function is vital, as Btk is essential for proper B cell maturation, and Btk deficiency can lead to severe immunodeficiencies such as X-linked agammaglobulinemia in humans. While Btk is primarily expressed in B cells, Sab shows broader distribution across various tissues, suggesting potential interactions with other protein kinases and influence on diverse signaling pathways. Sab monoclonal antibody (PL-A23) serves as an invaluable tool for researchers studying B cell signaling and regulatory networks governing immune responses.
