    Sab Antibody (A-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Sab protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Sab (A-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Sab protein plays a critical role as a Src homology 3 domain (SH3) binding protein, primarily associating with Bruton′s tyrosine kinase (Btk), which is essential for B cell development in both humans and mice. Btk is a member of the Btk/Tec family of cytoplasmic tyrosine kinases and is specifically crucial for B cell maturation; Btk deficiency leads to severe immunodeficiencies such as X-linked agammaglobulinemia in humans and X-linked immunodeficiency in mice. Sab acts as a negative regulator of Btk activity, and Sab binding to Btk not only reduces phosphorylation of Btk substrates but also inhibits Btk-induced auto-phosphorylation in B cells, thereby modulating immune responses. The interaction between SH3 domains of Sab and Btk is vital for this regulatory function, highlighting Sab′s importance in maintaining kinase activity balance within the immune system. Sab′s broader expression compared to Btk suggests potential interactions with additional protein kinases across various tissues, indicating possible roles in diverse cellular processes.
