RUFY1 Antibody (A-4) is a mouse monoclonal IgG2a kappa light chain antibody that detects RUFY1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). RUFY1 Antibody (A-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. RUFY1, also known as RUN and FYVE domain containing 1, RABIP4, or ZFYVE12, is a 708 amino acid protein that plays a crucial role in cellular processes by localizing to the cytoplasm and early endosome membrane. Such localization is vital for binding phosphatidylinositol 3-phosphate-containing vesicles, which are essential for early endosomal trafficking. RUFY1 contains one RUN domain and one FYVE-type zinc finger, enabling specific interaction with phosphatidylinositol 3-phosphate, thereby facilitating transport and sorting of endosomal cargo. RUFY1 undergoes post-translational modifications, including phosphorylation, which can occur upon DNA damage through ATM or ATR kinases. Phosphorylation of specific tyrosine residues, such as Tyr 389 and Tyr 400, is believed to be critical for proper localization to endosomes. Three isoforms of RUFY1 arise from alternative splicing, contributing to regulatory mechanisms and functional diversity in various tissues, including testis, lung, brain, and kidney.
