RPE65 Antibody (E-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects RPE65 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). RPE65 (E-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. RPE65, a member of the beta-carotene dioxygenase family, plays a crucial role in the visual cycle by facilitating the conversion of all-trans-retinyl esters to 11-cis-retinal, which is essential for the regeneration of visual pigments in photoreceptor cells. This process is vital for maintaining vision, as it allows for the proper functioning of the phototransduction cascade. RPE65 is primarily located in the retinal pigment epithelium, where RPE65 is anchored to the membrane via a lipid modification known as palmitoylation; this membrane-bound form is critical for enzymatic activity. Mutations in the RPE65 gene can lead to severe retinal diseases, including autosomal dominant retinitis pigmentosa and Leber congenital amaurosis type 2, highlighting the importance of RPE65 in ocular health and vision preservation.

How should the blocking peptide sc-390787 P be used?

Asked by: jenniferc
Thank you for your question. The blocking peptide is intended to be used as a negative control by preadsorbing the RPE65 (E-5): sc-390787 monoclonal antibody with it prior to application. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
