


Rootletin Antibody (E-5) is a mouse monoclonal IgG1 antibody that detects Rootletin in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Rootletin, also known as CROCC (ciliary rootlet coiled-coil protein), is a 2017 amino acid protein that plays a crucial role in the formation of centriole-associated fibrous structures, serving as an essential component of the ciliary rootlet. Rootletin is primarily localized to basal bodies and centrosomes in both ciliated and nonciliated cells, where Rootletin associates with the proximal ends of basal bodies. In photoreceptors, Rootletin is vital for forming elongated polymers between centrioles, which is important for maintaining the structural integrity and function of cilia. Additionally, Rootletin is required for centrosome cohesion and interacts with C-Nap1, a centrosomal protein that regulates the linkage of centrioles to basal bodies. Rootletin exists as a homopolymer, and Rootletin′s association with centrosomes can be modulated through phosphorylation by Nek2 (NIMA-related kinase 2), highlighting the significance of post-translational modifications in Rootletin′s function. Furthermore, two isoforms of Rootletin arise from alternative splicing events, adding another layer of complexity to Rootletin′s regulation and function in cellular processes.
