Rootletin Antibody (C-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects Rootletin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Rootletin antibody (C-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Rootletin, also known as CROCC (ciliary rootlet coiled-coil protein), is a 2017 amino acid protein that plays a crucial role in forming centriole-associated fibrous structures, serving as an essential component of the ciliary rootlet. Rootletin′s localization to basal bodies and centrosomes in both ciliated and nonciliated cells is vital for maintaining cellular organization and function. Specifically, Rootletin associates with proximal ends of basal bodies and forms elongated polymers between them in photoreceptors, which is critical for structural integrity and function of cilia. Furthermore, Rootletin is required for centrosome cohesion and interacts with C-Nap1, a centrosomal protein that regulates linkage of centrioles to basal bodies. Rootletin exists as a homopolymer, and association with centrosomes can be modulated through phosphorylation by Nek2 (NIMA-related kinase 2), highlighting importance of post-translational modifications in function. Additionally, two isoforms of Rootletin arise from alternative splicing events, further diversifying functional roles in cellular processes.
