RNF219 Antibody (A-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects RNF219 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). RNF219 (A-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. RNF219 protein, consisting of 726 amino acids, features a RING-type zinc finger motif, which is crucial for its role in the ubiquitination pathway, a key mechanism for protein degradation and cellular regulation. This motif allows RNF219 to bind two zinc ions, facilitating interaction with other proteins involved in transcriptional regulation and cellular response to DNA damage. Notably, RNF219 undergoes phosphorylation in response to DNA damage, likely mediated by ATM or ATR kinases, highlighting importance in maintaining genomic stability. The gene encoding RNF219 is located on human chromosome 13, a region that harbors over 400 genes, including critical tumor suppressor genes like BRCA2 and RB1. Understanding RNF219 function and interactions is vital, as dysregulation can contribute to various diseases, including cancer, making RNF219 a significant target for research and therapeutic development.

For Western Blot, is it recommended to use denatured or non-denatured conditions with RNF219 (A-4): sc-514812 monoclonal antibody?

Rated 1 out of 5 by from not goodIt didn't work for WB. I diluted it 1000:1, and incubated into overnight. My samples are HT-29 and L929 cell lysates. Also, when human HA tagged RNF219 was overexpressed on the 293T cell line, it was detected as HA specific antibody, but it did not work on RNF219 specific antibody.
