RNase H1 Antibody (H-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects RNase H1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). RNase H1 (H-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. RNase H1 plays a crucial role in the degradation of RNA within RNA-DNA hybrids, which is essential for maintaining genomic integrity and facilitating proper DNA replication and repair processes. This endonuclease activity is particularly important in cellular responses to DNA damage, as RNase H1 helps prevent the accumulation of potentially harmful RNA-DNA hybrids that can interfere with normal cellular functions. The human RNase H1 gene is located on chromosome 2p25.3, with pseudogenes found on chromosomes 17p11.2 and 1q, and RNase H1 is expressed at consistent levels across various human tissues, suggesting a function as a housekeeping protein vital for cellular homeostasis.

Why do the untransfected 293T cells (sample western blot data above, panel 3) show no RNase H expression? RNase H expression is ubiquitous.

Thank you for your question. We have tested a variety of different RNase H1 antibodies, including our mouse monoclonal, RNase H1 (H-4): sc-376326, in untransfected and transfected 293T cells and the results typically do not show a signal detected by Western blot in the untransfected 293T cell lysate. This could be for a variety of reasons. Although RNase H protein expression is ubiquitous, it may not be expressed abundantly enough in 293T to be detected by Western blot. Also, the purpose of our Western blot tests was to evaluate the antibody by identifying positive upregulation of the protein in the transfected cell lysate. Therefore, our Western blot conditions used were not optimized to detect signal in the untransfected cells. For example, the amount of protein loaded on the gels, our antibody incubation times, and the exposure times during the ECL reaction, all could be optimized if the goal was to detect endogenous protein in the untransfected control lysate.
