RICK Antibody (A-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects RICK protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-RICK antibody (A-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. RICK, also known as receptor interacting serine/threonine kinase 2 (RIPK2) or CARDIAK, plays a crucial role in regulating apoptosis and activating nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB). RICK protein is primarily located in the cytoplasm, where RICK interacts with various signaling complexes essential for mediating cellular responses to stress and inflammation. RICK′s localization allows rapid response to signals from tumor necrosis factor receptors (TNFR) and other apoptotic pathways, facilitating recruitment of downstream signaling molecules. RICK′s presence in these complexes is vital for transduction of apoptotic signals, and RICK dysregulation can lead to pathological conditions, including cancer and autoimmune diseases. Understanding RICK′s cellular dynamics and interactions provides insights into RICK′s role in immune responses and potential therapeutic targets for modulating apoptosis in various diseases.

Can RICK (A-10): sc-166765 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, RICK (A-10): sc-166765 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 4 out of 5 by from Nice endogenous Signal in Western Blot from HeLaThe antibody gives a nice specific signal of HeLa endogenous RIP2 in Western Blot. Unfortunately in my hands it did not give a nice cytoplasmic IF staining in PFA-fixed HeLa cells.

Rated 4 out of 5 by from Anti-RICK1 (RIPK2) AntibodyMouse anti-RICK1 antibody (sc-166765) works great on immunoblotting at 1:1000 in 2.5 % milk powder. It works well on IHC at 1:200 and can be reused for immunoblotting. It also works great on IP but not for in vitro kinase assays. It recognizes a 60 kDa band in all cell lines tested so far.

Rated 4 out of 5 by from Potent and quite specific antibodyThis monoclonal antibody was independently tested in-house by Kinexus Bioinformatics Corporation and validated for use for Western blotting. There was medium detection of the ~61 kDa target protein in a HeLa/Jurkat cell lysate mix, but not detected in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested. A ~46 kDa weak cross-reactive protein was evident in the HeLa/Jurkat cell lysate mix.
