Rho C Antibody (C-10) is a mouse monoclonal IgG kappa antibody specific for an epitope mapping between amino acids 158-189 at the C-terminus of human Rho C, reactive with mouse, rat, and human proteins, and suitable for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), enzyme-linked immunosorbent assay (ELISA), and immunohistochemistry. RhoC is a small GTPase belonging to the Rho family within the Ras superfamily, playing a pivotal role in regulating the actin cytoskeleton, cell shape, motility, and adhesion. RhoC (C-10) antibody localizes primarily in the cytoplasm and associates with the inner surface of the plasma membrane, which is crucial for signal transduction pathways that control various cellular processes. The membrane localization of RhoC is mediated by post-translational modifications at its C-terminus, particularly the addition of lipid groups to the CAAX motif, allowing interactions with membrane-associated proteins and influence on cytoskeletal dynamics. This cellular localization is important because RhoC′s activity at the plasma membrane is essential for regulating cell migration and invasion, processes that are often dysregulated in cancer. Overexpression or hyperactivation of RhoC has been associated with increased metastatic potential in several human cancers, including breast, pancreatic, and melanoma. Detected Rho C at expected size in 15 ug total proteine lysate, wet transfer one hour at 450 mAmps on PVDF membrane, overnight blocking with 5% nonfat dry milk in PBS, one hour incubation at RT with 1:500 anti Rho C in 5% PBS with 0,2%Tween followed by one hour incubation with 1:10.000 anti mouse HRP conjugated antibody and detection with ECL prime substrate followed by short film exposure (under 1 minute) and automatic processing.
