Rent1 Antibody (E-8) is a mouse monoclonal IgG2b antibody that detects Rent1 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Rent1, also known as the regulator of nonsense transcripts and HUpf1, plays a crucial role in the nonsense-mediated RNA decay (NMD) pathway, which maintains cellular integrity by preventing the translation of defective mRNA transcripts that could lead to harmful truncated proteins. Rent1 is characterized by an N-terminal zinc finger-like domain and NTPase domains, which classify Rent1 as a member of the superfamily group I helicase. Rent1 exhibits nucleic-acid-dependent ATPase activity and 5′ to 3′ helicase activity, essential for unwinding RNA during the decay process. Additionally, Rent1 functions as an RNA-binding protein, with activity modulated by ATP, and directly interacts with Rent2, a mammalian homolog of Upf2p, to facilitate the NMD process. Rent1 and interacting partners localize primarily in the cytoplasm, underscoring the importance of spatial dynamics in regulating RNA quality control mechanisms. The presence of Rent3a and Rent3b, mammalian orthologs of Upf3p, highlights the intricate network of protein interactions that link splicing and NMD, emphasizing Rent1′s critical role in safeguarding cellular function and embryonic viability.
