Rb Antibody (IF8) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Rb protein (also called retinoblastoma (RB) transcriptional corepressor 1 or RB1) of mouse, rat and human origin by western blotting, immunoprecipitation, immunofluorescence, immunohistochemistry on paraffin embedded sections, and flow cytometry. Rb monoclonal antibody (IF8) is available as both non-conjugated form, as well as multiple conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. Pediatric cancer retinoblastoma and formation of other human tumors can be attributed to mutations in the retinoblastoma tumor suppressor gene. Rb protein regulates differentiation, apoptosis and cell cycle control by coordinating the cell cycle, at G1/S, with transcriptional machinery that includes the heterodimeric E2F family. During G1, cyclin D (D1, D2, D3)-dependent kinase-mediated phosphorylation of Rb at Ser 795 marks the conversion of Rb from a transcriptionally repressive, hypophosphorylated state to an inactive, phosphorylated state, which may be sustained through mitosis by differential phosphorylation of up to 16 putative serine or threonine residues, including Ser 249/Thr 252, Thr 373, Thr 356, Ser 780, Ser 807/Ser 811 and Thr 821/Thr 826. Hypophosphorylated Rb represses transcription of genes controlling cell cycle through direct protein-protein interactions, by binding and inactivating promoters of transcription factors, and through recruitment of histone deacetylase, which deacetylates promoter regions and enhances nucleosome formation, thereby masking transcription enhancing cis elements. Anti-Rb monoclonal antibody (IF8) is recommended for researchers investigating cell cycle regulation and molecular mechanisms underlying cancer development.

Do you happen to know whether this mAb (IF8) is reactive with pig (porcine) RB by IHC and WB, looking for a pig RB antibody? Thanks!

Thank you for your question. Rb Antibody (IF8): sc-102 is recommended for detection of Rb p110 of mouse, rat and human origin by WB, IP, IF, IHC(P) and FCM. We have no internal data for this antibody's reactivity in porcine tissues and it is not recommended for this use. However there is a publication in which this antibody was used for detection of Rb in pig. The corresponding PMID is 16293970.

Hello My name is Laura, I bought the antibody sc-102 Rb and I am getting two bands, I need to know... ¿Is it a phosphorylated form of Rb or a nonspecific band? thank you so much

Thank you for your question. We would recommend treating your lysates with Lambda Phosphatase: sc-200312A to confirm whether or not the antibody is specifically detecting phosphorylated Rb in your samples.

Does this antibody (SC_102) bind with other proteins in Rb family? I can detect three bands of which their M.W. are within range of protein marker from 100 to 150 kD. Are all these bands Rb protein? (protein lysate from MCF7, 293T; Ab dilution 1:800)

Thank you for your question. This mouse monoclonal antibody, Rb (IF8): sc-102, is specific for the detection of Rb. It sounds like your results are inconsistent with our Western blot results published on our website for this antibody. We would recommend you include a secondary antibody control (omit primary antibody incubation) to confirm if any of the other bands you are seeing are due to non-specific binding of the secondary antibody alone. Does this "Rb Antibody (IF8)" bind with both the phosphorylated and the un-phosphorylated Rb proteins? I can then detect the relative number of the phosphorylated Rb with one of the p-Rb antibodies, is this correct? Thanks.

Thank you for your question. This mouse monoclonal antibody, Rb (IF8): sc-102, is not phospho-specific and therefore should be able to detect both the un-phosphorylated and phosphorylated Rb protein. If you use this antibody as a control along with a phospho-specific Rb antibody you will observe a qualitative but not a quantitative difference by Western blot.

The band is specific but very weak. The antibody was diluted 500 times into 5% BSA, into TBST and incubated over-night at 4°C with the membrane.
