Rb Antibody (C-2) is a mouse monoclonal IgG2a kappa light chain antibody raised against amino acids 769-921 at the C-terminus of mouse retinoblastoma protein (Rb), a crucial cell cycle regulator and tumor suppressor. Rb mouse monoclonal antibody (C-2) recognizes an epitope within the specified immunogen sequence and demonstrates broad species cross-reactivity, detecting Rb in mouse, rat, and human samples through Western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) applications. Anti-Rb monoclonal antibody (C-2) is available in non-conjugated form and multiple conjugated forms including agarose, HRP, PE, FITC, and various Alexa Fluor® conjugates, offering versatility for diverse experimental needs. The retinoblastoma protein, encoded by the RB1 gene on chromosome 13q14.2, undergoes complex regulation through phosphorylation to control cell cycle progression, differentiation, and apoptosis. In hypophosphorylated form, Rb binds to E2F transcription factors, actively repressing genes required for S-phase entry. Multiple phosphorylation events by cyclin D-dependent kinases at specific residues (Ser249/Thr252, Thr373, Thr356, Ser780, Ser795, Ser807/Ser811, and Thr821/Thr826) trigger Rb inactivation and E2F release, promoting cell cycle advancement. Beyond direct transcriptional control, Rb recruits histone deacetylase complexes to target promoters, inducing chromatin condensation and enhancing transcriptional repression. The RB1 gene produces multiple splice variants, generating various Rb isoforms with potentially distinct functions based on their domain composition and ability to interact with regulatory partners. Mutations or dysregulation of RB1 can compromise Rb′s tumor suppressor functions, contributing to retinoblastoma and other human cancers. Through these diverse cellular roles, Rb (C-2) antibody serves as a valuable tool for investigating cell cycle regulation, differentiation, apoptosis, and tumorigenesis mechanisms.
