RANKL Antibody (G-1) is a mouse monoclonal IgG1 kappa kappa light chain antibody raised against amino acids 46-317, representing full-length RANKL of human origin, effectively detecting RANKL proteins of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). RANKL, also known as Receptor Activator of Nuclear Factor Kappa-B Ligand, TNFSF11, CD254, TRANCE, OPGL, or ODF, is a type II transmembrane protein crucial for bone remodeling and immune system regulation. Post-translational modifications of RANKL, such as glycosylation, are important for stability, activity, and interaction with RANK receptor. Glycosylation enhances RANKL′s ability to bind to RANK, activating signaling pathways like NF-κB and JNK, which are essential for osteoclast differentiation and activation. The structural integrity and post-translational modifications of RANKL are vital for function in regulating T cells and osteoclasts, impacting processes like bone resorption and immune responses. Alterations in RANKL expression or modifications can contribute to pathological conditions such as osteoporosis, rheumatoid arthritis, and bone metastasis. RANKL monoclonal antibody (G-1) offers high specificity and sensitivity for detecting RANKL, available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Anti-RANKL antibody (G-1) serves as an essential tool for researchers studying the role of RANKL in bone metabolism and immune regulation, aiding in the development of therapeutic interventions for related diseases.

Hello! I purshed conjugate antibody aganist RANKL 377079HRP in order to detect RANKL in a bone sample by IHC(P) and I followed the immunoperoxidase staining protocol that you provide on-line. However, the processing is not working. Can you help me?

Does it work well on primary cell lysates?

Thank you for your question. RANKL Antibody (G-1): sc-377079 is recommended for detection of RANKL of mouse, rat and human origin by WB, IP, IF and ELISA. This recommendation is valid for lysates generated from both cell cultures and tissue material.

Boa Tarde! Nos comentários vocês não indicam IHQ para bloco parafinado, porém nas especificações conta isso "disponível conjugado a agarose para IP; HRP para WB, IHC (P) e ELISA; e para ou ficoeritrina ou FITC para IF, IHC (P) e FCM" Estou com duvidas!

Thank you for your question. The conjugated form of this antibody is recommended for IHC on paraffin sections, however, the non-conjugated form has been warrantied for IF on fresh frozen tissue sections.

Which is the actual molecular weight of RANKL? the data sheet says 40-35 Kd, but I have got bands around 50Kd when using primary clavaria cells from mice...

The expected molecular weight of RANKL, as detected by RANKL Antibody (G-1): sc-377079, depends on the form present in your sample: RANKL full-length (35-40 kDa), membrane-bound RANKL (34-40 kDa), or soluble RANKL (20-30 kDa) . What's the difference between the sc-52950 and the sc-377079 HRP in IHC with mouse tissue? And what about the result?Will they be same?

Both sc-52950 and sc-377079 are mouse monoclonal antibodies recommended for detection of RANKL of mouse, rat and human origin. sc-52950 can be used on paraffin-embedded tissues, while sc-377079 is not recommended for such application. sc-377079 HRP is sc-377079 directly conjugated to HRP. When using sc-377079 HRP in WB or IHC, the secondary reagent can be avoided. How much protein do you recommend loading on the gel for Western blot detection using RANKL (G-1): sc-377079 monoclonal antibody?
