Pygopus 2 Antibody (B-8) is a mouse monoclonal IgG1 antibody that detects Pygopus 2 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Pygopus 2 is a 406 amino acid protein that serves as the human homolog of the Drosophila pygopus protein and is primarily localized to the nucleus, where Pygopus 2 plays a pivotal role in the Wnt signaling pathway. Pygopus 2 contains a PHD finger that facilitates interaction with the homology domain of the Wnt signaling protein Bcl-9, effectively linking Pygopus 2 to the β-catenin/TCF complex, which is essential for the transcriptional activation of Wnt target genes. Proper functioning of Pygopus 2 is critical for regulating signal transduction within the Wnt pathway; disruptions in the Pygopus 2/β-catenin interaction have been implicated in the development of B-cell malignancies.
