Pygopus 2 Antibody (B-12) is a mouse monoclonal IgG2a kappa light chain antibody that detects Pygopus 2 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Pygopus 2 antibody (B-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Pygopus 2, also known as PYGO2, is a 406 amino acid protein that serves as the human homolog of the Drosophila pygopus protein. Pygopus 2 is primarily localized in the nucleus, where Pygopus 2 plays a pivotal role in the Wnt signaling pathway. Pygopus 2 contains a PHD finger that facilitates interaction with the homology domain of the Wnt signaling protein Bcl-9, effectively linking Pygopus 2 to the β-catenin/TCF complex, which is essential for the transcriptional activation of Wnt target genes. Proper functioning of Pygopus 2 is crucial for regulating signal transduction within this pathway; disruptions in the Pygopus 2/β-catenin interaction have been implicated in the development of B-cell malignancies. Pygopus 2 expression is notably upregulated in breast cancer cells, indicating a significant role in carcinogenesis and suggesting potential as a target for therapeutic intervention in cancer treatment.
