PTH Antibody (2B67) is a mouse monoclonal IgG1 antibody that detects parathyroid hormone (PTH) in human, bovine, and canine samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. PTH is a crucial 84 amino acid peptide that plays a vital role in regulating calcium metabolism by increasing blood calcium levels through multiple mechanisms, such as stimulating bone formation, which enhances bone mass and strength in both rats and humans. The functional significance of PTH is primarily attributed to the first 34 amino acids at its amino terminus, which are essential for its biological activity. The gene encoding PTH is located on human chromosome 11p15.3-p15.1, and parathyroid hormone-related protein (PTH-rP), which shares structural similarities with PTH, is produced by various cell types and is involved in regulating endochondral bone development and epithelial-mesenchymal interactions during mammary gland and tooth formation. PTH-rP, isolated from a human lung cancer cell line, exhibits PTH-like effects that can lead to humoral hypercalcemia of malignancy. Both PTH and PTH-rP are influenced by vitamin D and steroid hormones, and preferentially bind to specific PTH/PTH-rP receptors, activating adenylate cyclase or phospholipase C beta through protein kinase C activation.
