PTEN Antibody (F-1) is a mouse monoclonal IgG1 kappa light chain antibody specifically designed to recognize the N-terminal epitope of PTEN, also known as phosphatase and tensin homolog, MMAC1, or TEP1. Anti-PTEN antibody (F-1) targets amino acids 3-29 of human PTEN and is validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) across a wide range of species, including mouse, rat, human, equine, canine, bovine, porcine, and avian origins. PTEN is a critical tumor suppressor protein that plays a vital role in regulating cell growth, proliferation, and survival by dephosphorylating phosphatidylinositol (3,4,5)-trisphosphate, thereby antagonizing the PI3K/AKT signaling pathway. Structurally, PTEN consists of an N-terminal phosphatase domain and a C-terminal C2 domain, which are essential for enzymatic activity and membrane binding, respectively. The integrity of these structural domains is crucial for PTEN′s ability to maintain cellular homeostasis and prevent uncontrolled cell division, making proper function indispensable in preventing tumorigenesis. Additionally, PTEN interacts with various proteins and cellular membranes, facilitating a role in multiple signaling pathways that suppress tumor growth. Mutations or loss of PTEN disrupt structural configuration, leading to impaired function and contributing to the development of numerous cancers, including glioblastomas, breast, kidney, and prostate cancers.
