
PRP8 Antibody (F-6) is a mouse monoclonal IgG1 antibody that detects PRP8 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PRP8, also known as pre-mRNA-processing-splicing factor 8, is a highly conserved nuclear protein that serves as a central component of the spliceosome′s catalytic core, playing a crucial role in pre-mRNA splicing. PRP8 is essential for transesterification reactions that facilitate spliceosome-induced pre-mRNA splicing, particularly through interaction with the GU dinucleotide at the 5′ splice site, forming a specific UV-inducible cross-link. PRP8′s functional interaction with the 3′ splice site significantly influences the efficiency of the second catalytic step in splicing. PRP8 is involved in the first transesterification step, as mutations that disrupt the negative regulation of PRP28 or PRP44/Brr2 can block U4 activation, highlighting PRP8′s critical role in spliceosome assembly and function. PRP8 interacts with a conserved region of U6, which is vital for the formation of the spliceosome′s catalytic core, underscoring PRP8′s importance in gene expression regulation.
