I am processing Chromatin samples from mouse liver hepatocytes. I would like to know what would be the recommended dilution? I did not purchase the Trans Cruz antibody so starting with a diluted concentration.

The antibody (PPARα Antibody (H-2) HRP) is already conjugated to the peroxidase?! So, it does not need to the secondary antibody for Western Blot analysis, right?. Thank you.

Yes, that is correct. sc-398394 HRP is directly conjugated to HRP enzyme. There is no need to use any secondary antibody for detection.

I work with U87 MG cells and I´d used this antibody but I have many unspecific bands. The image in the datasheet shows two bands for that type of cells, at 55 k and at 90k. If PPAR is supose to be the one at 55, is the one at 99 and isoform?

Will this antibody work with a regular secondary Anti-mouse IgG, HRP-linked Antibody? Or is it necessary to get the recommended secondary? Will there be non-specific bands?

Thank you for your question. We do recommend the Mouse IgG Binding Protein over conventional anti-mouse secondaries as we find them to be more specific and consistent, however the primary antibody will work with conventional anti-mouse secondaries as well.

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, PPARα (H-2): sc-398394 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Hello, To those that have managed to get this to work on HepG2 cells, can you please share the WB protocol? I have not been able to get it to work on a Western

Thank you for your question. I have provided a link below to our recommended protocol for Western blotting with this mouse monoclonal antibody, PPARα (H-2): sc-398394. We do not have any published data in Hep G2 for PPARα. According to the human protein atlas, (http://www.proteinatlas.org/ENSG00000186951-PPARA/cell), although there is verified RNA expression of PPARα in Hep G2, they failed to detect the protein using an antibody using immunofluorescent staining. We recommend using HUV-EC-C (sc-364180) and KNRK (sc-2214) whole cell lysates as positive controls with this antibody. We loaded 50 µg whole cell lysate protein, and used the primary antibody at a dilution of 1:200 incubated for one hour at room temperature in Blotto (sc-2333). Please contact our Technical Service Department for additional support in troubleshooting this antibody.
