Positive cofactor 4 Antibody (H-12) is a mouse monoclonal IgG2a kappa light chain antibody that detects Positive cofactor 4 (PC4) of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PC4 Antibody (H-12) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PC4 plays a crucial role in transcription regulation within eukaryotic cells as part of high molecular weight coactivating complexes that facilitate communication between transcriptional activators and RNA polymerase II (RNAPII). PC4 interacts with transcription factor IIA (TFIIA) and TATA-binding protein-associated factors (TAFs), enabling direct binding to double-stranded DNA. This interaction is essential for activating and repressing RNAPII-mediated transcription, varying with specific transcription factors and holoenzyme components. PC4 undergoes phosphorylation by TFIID and TFIIH, leading to PC4 release from the DNA promoter region, inhibiting assembly into the transcriptional promoting complex and blocking transcription. Such dynamic regulation underscores PC4′s importance in gene expression and cellular function, making PC4 a valuable target for research in transcriptional control mechanisms.

When should the TransCruz reagent sc-166280 X be used instead of the standard form sc-166280?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
