Pol II Antibody (mara-3) is a mouse monoclonal IgM antibody that detects RNA polymerase II (Pol II) in mouse, rat, and human samples through western blotting (WB) and immunoprecipitation (IP) applications. Pol II is a crucial enzyme composed of twelve subunits, responsible for the transcription of protein-coding genes, which is a fundamental process for gene expression and cellular function. The largest subunit of Pol II, known as RPB1 or RPB205, is a 1,840 amino acid protein that features a C2H2-type zinc finger and a C-terminal domain rich in heptapeptide repeats. This subunit is essential for catalytic activity and plays a vital role in forming the DNA-binding domain, necessary for accurate transcription of the DNA template. RPB1′s proper function is critical; any disruption severely compromises mRNA synthesis and transcription elongation, potentially disrupting cellular processes and gene regulation. Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
