Podocin Antibody (G-5) is a mouse monoclonal IgM antibody that detects Podocin protein of mouse and rat origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Podocin (G-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Podocin plays a crucial role in maintaining the integrity of the glomerular filtration barrier, which is essential for proper kidney function. Podocin is primarily located in the podocytes of the kidney, where Podocin contributes to the formation of the slit diaphragm, a structure that regulates the passage of proteins and other molecules from the blood into the urine. Disruption of Podocin function can lead to severe kidney disorders, such as autosomal recessive steroid-resistant nephrotic syndrome (SRN1), characterized by significant proteinuria and rapid progression to end-stage renal disease. The human NPHS2 gene, which encodes Podocin, is located on chromosome 1q25-q31 and produces a 383 amino acid protein that exhibits a unique hairpin-like structure, with both amino- and carboxy-termini situated intracellularly. This structural configuration, along with homology to stomatin family proteins, suggests that Podocin may form high-order oligomers that serve a scaffolding function, thereby influencing the stability and functionality of the glomerular filtration barrier. Understanding Podocin′s role in kidney health is vital for developing targeted therapies for related diseases.
