Podocalyxin-like 1 Antibody (B-11) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 23–322 within the N-terminal extracellular domain of human Podocalyxin-like protein 1 (PODXL). Also known as PCLP1 or simply podocalyxin, PODXL is a transmembrane sialomucin characterized by a heavily glycosylated extracellular domain, a single-pass transmembrane region, and a cytoplasmic tail containing multiple phosphorylation sites. PODXL′s structure is crucial because its extensive N- and O-linked glycosylation and sialylation confer a strong negative charge, which imparts anti-adhesive properties essential for maintaining the foot processes of podocytes in the glomerular filtration barrier of the kidneys. PODXL is also expressed in various epithelial tissues, including the breast, pancreas, and prostate, in addition to endothelial cells and hemangioblasts, precursors to hematopoietic stem cells (HSCs), making PODXL a valuable marker for hemangioblast detection and studies of epithelial biology. PODXL is involved in cancer progression and metastasis, particularly in breast and prostate cancers, highlighting PODXL′s importance in oncological research. PODXL interacts with the actin cytoskeleton through association with ezrin, which is essential for maintaining cell shape and polarity, and serves as a ligand for L-selectin, facilitating the rolling of leukocytes on endothelial cells during the inflammatory response. Structurally related to CD34, PODXL plays a significant role in mediating interactions between leukocytes and endothelial cells during inflammation.
