
Podocalyxin-like 1 Antibody (3D3) is a mouse monoclonal IgG1 kappa light chain antibody raised against human recombinant podocalyxin-like protein, specifically detecting Podocalyxin-like protein 1 (PODXL) of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Also known as PCLP1, or simply podocalyxin, PODXL is a transmembrane sialomucin characterized by extensive post-translational modifications, including heavy N- and O-linked glycosylation and sialylation of its extracellular domain. These modifications confer a strong negative charge, contributing to PODXL′s anti-adhesive properties and maintaining the podocyte foot processes essential for the glomerular filtration barrier in the kidneys. The intracellular domain of PODXL contains multiple phosphorylation sites, which play a significant role in intracellular signaling pathways that regulate cell adhesion, migration, and polarity. Beyond the kidneys, PODXL is expressed in various epithelial tissues, including the breast, pancreas, and prostate, as well as in endothelial cells and hemangioblasts, precursors to hematopoietic stem cells. PODXL is involved in cancer progression and metastasis, particularly in breast and prostate cancers, underscoring its clinical relevance in oncology. PODXL interacts with the actin cytoskeleton through association with ezrin, which is essential for maintaining cell shape and polarity, and serves as a ligand for L-selectin, facilitating leukocyte rolling on endothelial cells during inflammatory response. Structurally related to CD34, PODXL mediates interactions between leukocytes and endothelial cells during inflammatory processes. Available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, anti-PODXL antibody (3D3) is a versatile tool for researchers investigating PODXL′s role in physiological and pathological conditions, such as cancer progression and metastasis due to its influence on cell adhesion and migration.
