

PNPase Antibody (C-5) is a mouse monoclonal IgG1 antibody that detects PNPase in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PNPase, also known as mitochondrial polyribonucleotide nucleotidyltransferase, 3′-5′ RNA exonuclease, OLD35, or PNPT1, plays a crucial role in mRNA degradation by hydrolyzing single-stranded ribonucleotides in the 3′ to 5′ direction, which is essential for maintaining cellular RNA homeostasis and regulating gene expression. PNPase is primarily located in the mitochondria, where PNPase forms homotrimers and is upregulated in response to interferon-β induction, highlighting PNPase′s involvement in cellular stress responses. The N-terminus of PNPase contains a putative mitochondrial targeting sequence, and mutation analysis has confirmed that these sequences are critical for directing PNPase to the mitochondria, ensuring proper localization and function. Additionally, endogenous PNPase has been shown to co-localize with mitochondrial marker proteins in HeLa cells, further emphasizing PNPase′s importance in mitochondrial RNA metabolism and overall cellular health.
