PMCA1/4 Antibody (5F10) is a mouse monoclonal IgG2a kappa light chain antibody that detects PMCA1/4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-PMCA1/4 antibody (5F10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The plasma membrane Ca2+-pumping ATPase (PMCA) family consists of four genes, PMCA1-4, with PMCA4b being predominantly expressed in developing mammary tissue. During lactation, PMCA1b expression increases while PMCA4b expression decreases, highlighting the critical role of PMCAs in maintaining cellular calcium homeostasis. Notably, PMCA4b undergoes cleavage at Asp 1080 by caspase-3, resulting in a fully active fragment that responds more rapidly to increases in calcium levels compared to its autoinhibited form. This rapid response is essential for regulating intracellular calcium levels, particularly in apoptotic cells, where PMCA4b may play a significant role. Additionally, PMCA1 and PMCA4 are expressed at comparable levels in astrocytes and neurons, indicating their importance in neuronal function and signaling.

I am using PMCA1/4 (5F10): sc-20028 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
