PIWI Antibody (G-1) is a mouse monoclonal IgG2b kappa light chain antibody that detects PIWI protein of Drosophila melanogaster origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-PIWI antibody (G-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PIWI protein is a crucial component of the PIWI family, which is evolutionarily conserved and plays a significant role in stem cell self-renewal, gametogenesis, and RNA interference across organisms. PIWI protein is predominantly located in the nucleoplasm of both somatic and germline cells, where PIWI presence is vital for maintaining stem cell population balance. In the Drosophila ovary, PIWI protein is essential for asymmetric division of germline stem cells (GSCs), ensuring production and maintenance of daughter GSCs, although PIWI protein is not required for differentiation of committed daughter cells. The highly conserved C-terminal region of PIWI protein is critical for function, and removal from single GSCs significantly impairs division rate, indicating PIWI protein acts as a cell-autonomous promoter of GSC division. PIWI protein overexpression in somatic cells leads to increased GSC numbers and division rates, highlighting PIWI protein′s dual role as a stem cell dynamics regulator. PIWI (G-1) antibody serves as a valuable tool for studying stem cell regulation mechanisms and broader implications in developmental biology.
