PI-9 Antibody (F-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects PI-9 of mouse origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PI-9 (F-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Proteinase inhibitor-9 (PI-9), also known as cytoplasmic antiproteinase 3 (CAP3), is a crucial member of the serpin family, specifically the Ovalbumin family, and plays a significant role in regulating serine proteinase activity, particularly granzyme B, which is vital for controlling apoptosis in cytotoxic lymphocytes. The inhibition of granzyme B by PI-9 is particularly important as PI-9 prevents unwanted cell death during immune responses, thereby maintaining tissue integrity and function. PI-9 is predominantly expressed in the placenta, lung, and cytotoxic lymphocytes, and PI-9 presence in the nuclei of human cytotoxic cells, endothelial cells, and epithelial cells highlights PI-9′s role in cellular protection against apoptosis. Notably, PI-9 is rapidly induced by estrogen and upregulated in response to inflammatory stimuli, which further enhances PI-9′s protective role during target cell killing. Additionally, PI-9 expression has been observed in various human and murine tumors, indicating PI-9′s potential relevance in cancer biology and therapy.
