PF-4 Antibody (D-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects PF-4 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PF-4 (D-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Platelet factor-4 (PF-4) is a 70 amino acid protein released from the alpha-granules of activated platelets, playing a crucial role in hemostasis by binding with high affinity to heparin and related sulfated glycosaminoglycans. This interaction is vital as PF-4 neutralizes heparin-like molecules on the endothelial surface of blood vessels, thereby inhibiting local antithrombin III activity and promoting coagulation, which is essential for preventing excessive bleeding during vascular injury. Additionally, PF-4 acts as a potent chemoattractant for neutrophils and fibroblasts, indicating involvement in inflammation and wound healing processes. Studies have shown that PF-4 and eotaxin, a specific chemoattractant for eosinophils, exhibit increased expression in the spleens of adult NOA mice, an animal model for allergic dermatitis, suggesting a potential link between PF-4 and the pathophysiology of atopic dermatitis. PF-4 is classified as a small inducible gene (SIG) due to its small size and upregulation upon platelet activation, mapping to human chromosome 4q13.3.
