PDPc Antibody (D-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects PDPc protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PDPc (D-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The pyruvate dehydrogenase phosphatase (PDP) is a crucial serine phosphatase that plays a vital role in cellular metabolism by catalyzing the dephosphorylation and reactivation of the alpha subunit of the E1 component of the mitochondrial pyruvate dehydrogenase multienzyme complex. PDP is composed of a heterodimer of catalytic and regulatory subunits, with PDPc (pyruvate dehydrogenase phosphatase, catalytic subunit 1), a 537 amino acid protein localized within the mitochondrial matrix, being particularly significant. PDPc efficiently dephosphorylates all three phosphorylation sites on the alpha chain of the E1 component, thereby activating pyruvate dehydrogenase to facilitate the conversion of pyruvate to acetyl-CoA, a critical substrate for the Krebs cycle. Additionally, PDPc is stimulated by calcium binding and requires two magnesium ions as cofactors for enzymatic activity. Defects in the gene encoding PDPc can lead to pyruvate dehydrogenase phosphatase deficiency, resulting in serious metabolic disorders such as lactic acidosis and neurological dysfunction, underscoring PDPc′s essential role in maintaining metabolic homeostasis.
