PDGFR-β Antibody (18A2) is a mouse monoclonal IgG1 kappa light chain antibody that detects PDGF receptor type β of human origin by immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). PDGFR-β monoclonal antibody (18A2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The platelet-derived growth factor (PDGF) receptor type β plays a crucial role in cell signaling, particularly in the regulation of cell growth, survival, and migration, which are vital processes in tissue repair and development. PDGFR-β is primarily located on the surface of mesenchymal and glial cells, where PDGFR-β mediates the effects of its ligand, PDGF-BB, leading to receptor dimerization and activation of downstream signaling pathways. PDGFR-β′s ability to bind specifically to the B subunit of PDGF is essential for its function, as PDGFR-β triggers a cascade of intracellular events that influence cellular behavior. Dysregulation of PDGFR-β signaling has been implicated in various pathologies, including chronic myelomonocytic leukemia (CMML), highlighting PDGFR-β′s importance as a therapeutic target in cancer treatment.

We recommend using sodium citrate buffer (pH 6.0) and heat treatment for antigen retrieval on formalin-fixed paraffin-embedded tissue sections.

Can PDGFR-β (18A2): sc-19995 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, PDGFR-β (18A2): sc-19995 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Antigen retrieval methods were optimized between without antigen retrieval, heat-induced citrate buffer, and trypsin-induced antigen retrieval. However, a positive signal was not clear on each slide.

Rated 5 out of 5 by from Great results for Flow citometryI received this antibody as a free sample. I used it for FACS and it worked very well. I worked with HFF(Human Foreskin Fibroblast) and Primary Breast Cancer Cells. First I tried with a dilution of 1ul per 100000 cells in HFF and the result wasn´t the expected. After, I used a dilution of 2ul per 100000 cells of HFF and 2ul per 200000 Primary Breast Cancer Cells. This time I obteined better results.
