SEE ALSO...

PDGFR-β Antibody (D-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects PDGFR-beta protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PDGFR-β (D-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Platelet-derived growth factor receptor beta (PDGFR-β) plays a crucial role in cell signaling pathways that regulate cell proliferation, survival, and migration, particularly in mesenchymal and glial cells. PDGFR-β is primarily located on the cell surface, where PDGFR-β interacts with platelet-derived growth factor (PDGF) to initiate a cascade of intracellular signaling events. PDGFR-β′s location enables responses to extracellular signals, vital for processes such as wound healing, tissue repair, and embryonic development. Dysregulation of PDGFR-β signaling has been implicated in various pathological conditions, including cancer and fibrotic diseases, highlighting PDGFR-β′s significance as a therapeutic target. PDGFR-β′s interaction with specific ligands, such as PDGF-BB, leads to receptor dimerization and activation of intrinsic tyrosine kinase activity, further emphasizing PDGFR-β′s cellular localization in mediating biological responses.

Do you have any recommendations of how to use PDGFRB D-6: sc-374573 in immunohistochemistry? We have tried it with Citrate Buffer and the ABC/DAB kits and it works well, although in some areas the staining looks weak. Any specific products needed?

Asked by: Sabine79
Thank you for your question. A link to our recommended protocol for use with this PDGFR-β Antibody (D-6): sc-374573 is provided below. We recommend using sodium citrate buffer (pH 6) with heat treatment for antigen retrieval. And we recommend using the primary at a starting dilution of 1:50. https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 1 out of 5 by from Not valid for IF on rat frozen liver sectionsRat liver frozen sections were subjected to immunofluorescence using PDGFR-β (D-6) antibody. Antigen retrieval methods were optimized between without antigen retrieval, heat-induced citrate buffer, and trypsin-induced antigen retrieval. However, a positive signal was not clear on each slide.
